Application: heparin can be combined with anticoagulant factor III, coagulation factor, protein synthesis factor, lipoprotein, interferon, nucleic acid binding protein, restriction endonuclease, thrombin and thrombin like biological macromolecule, so heparin agarose gel can be used for the purification of these substances.
Purpose: heparin is an acidic polysaccharide containing sulphate, which is coupled to cross-linked and activated agarose gel, which has high physical and chemical stability. Heparin and coagulation factors and other plasma proteins, lipoproteins, protein synthesis factors act on nucleic acids and steroid receptor enzymes, so heparin agarose gel can be used for purification of such substances.
Storage: sealed at 4-8 ℃. The used filler shall be thoroughly washed with pure water, and finally stored in 20% ethanol at 4 ℃.
matters needing attention:
(1) Before loading the sample, the sample must be filtered through the membrane and the pigment must be removed, otherwise the impurities and pigment will be adsorbed on the filler and affect the normal use of the filler. All buffers need to be filtered with a 0.45um filter.
(2) In the process of use, avoid using strong acids and bases with high concentration, and the concentration of acids and bases shall be less than 0.1 mol. Alkali slows the flow rate.
(3) Different samples have different adsorption and elution methods, which can be carried out according to the relevant literature.
